Abstract

This study was conducted during the period from 2016 to 2019 in the Central Laboratory for Date Palm Research and Development, Giza; Egypt. This study aimed at conducting a set of e xperiments for improving the growth of tissue culture plantlets of date palm cv. Bartamoda, during the rooting stage and obtaining a vigor plantlets, improving their characteristics in order to increase their success percentage during the acclimatization stage and reducing the loss percentage in the acclimatized plantlets. Shoots elongation and roots formation of shoots clusters or plantlets have been discussed by some e xperiments. Expe riment (1) addressed the effect of osmotic stress by sorbitol and mannitol sugars atdifferent concentrations (0.0, 0.1, 0.3, and 0.5 M) supplemented in the rooting medium on the recovery capacity of vegetative growth of date palm shoot clusters, best result was obtained with sorbitol sugar at 0.3 M for shoot elongation and good root system formation; also, chemical analysis after stress period then after recovery of explants was conducted. Experiment (2) addressed the effect of auxins and cytokinins balance on shoots elongation and roots formation of shoots clusters for three subculture, two auxins (NAA or IBA) at different concentrations (0.1 mg/ l and 0.5 mg/ l) in combination with three cytokinins (BA, Kin or 2IP) at constant concentration at (0.1 mg/l) supplemented in culture nutrient media, the best results for shoot elongation and root system formation were recorded at treatment of (0.5 NAA + 0.1 2ip). Expe riment (3) addressed the effect of the addition of different types of silicates (sodium silicate, potassium silicate and calcium silicate) at concentrations (0.25, 0.5, 1 g/l) supplemented separately in culture nutrient media for 8 weeks on shoots clusters elongation and root system formation, the best result was recorded with potassium silicate at a concentration was 1 g/l gave the best shoot elongation and good root system
