Abstract

Insomnia is a general complaint of poor sleep quality or inadequate quantity of sleep that is subjective and that may adversely affect daily functioning of individuals. People with insomnia may suffer one or several sleep disturbances that include; difficulty initiating sleep or difficulty maintaining sleep. These difficulties may result in personal problems during the day such as fatigue, diminished concentration, memory difficulty and inability to perform many tasks. Zaleplon (Z), an oral pyrrazolo pyrimidine hypnotic drug, belonging to the non-benzodiazepine so called {u2018}Z drugs{u2019} is used in the treatment of insomnia. (Z) is rapidly absorbed after oral administration but has very poor aqueous solubility. It undergoes extensive first pass hepatic metabolism with only 30% of the drug being systemically available. This makes its absorption dissolution rate limited and so delays its onset of action. The work in this thesis is divided into three chapters: Chapter I: Optimization of zaleplon dissolution enhancement through adsorption on bioceramic nanocarriers. Chapter II: Optimization of zaleplon dissolution enhancement using nanocrystallization technique. Chapter III: Pharmacodynamic and pharmacokinetic evaluation of intranasal zaleplon nanopowders
