Abstract

VII- Summary and Conclusion The present study was performed to investigate the role of oxytocin hormone in diabetes and obesity This study was carried on 173 Male Sprague-Dawley rats aged between 8 and 12 weeks old and the following experiments were carried out: Experiment 1: Effect of gastric gavage of Sweet condensed milk (SCM)/Cream on plasma oxytocin level in normal rats. 18 male SD rats were used and divided into 3 groups (6 rats of each): 4- Sweetened condensed milk (SCM) gavage group “SCM gavage”: 5 mL of SCM was infused slowly (140 µl /min) using infusion pump into the stomach via gavage tube. 5- Cream gavage group “cream gavage”: 5 mL of single cream was infused slowly (140 µl /min) using infusion pump into the stomach via gavage tube. 6- Sham operated control group “sham gavage”: In which nothing was infused. All rats have been fasted overnight for 16 hours, deeply anaesthetized, then three blood samples were withdrawn via femoral artery -5’,20’,40’ following start of gavage. Oxytocin measured in plasma by RIA. Experiment 2: Evaluation of phosphorylated extracellular receptor kinase (p-ERK) activation following voluntary consumption of sweet condensed milk.20 rats were used and divided into two groups (10 rats of each)::3- Sweet Condensed Milk group (SCM group): Rats were habituated to empty feeding glass bowl in their cage for 3 days. For the next 3 days, rats were habituated for 15 minutes access of 5ml SCM in glass bowls at specified time schedule. On the 7th day, rats were transcardially perfused with paraformaldehyde 30 minutes after the start of voluntary SCM consumption and brains were dissected. 4- Control group: On which the same procedure was done, however glass bowels were empty.
