Abstract

Cancer cachexia is a complex syndrome; it is most commonly seen in patients with gastrointestinal, lung, and prostate cancers. Cachexia causes reduced quality of life and shortened life expectancy. Although the mechanism of cancer cachexia is not well known, several key mediators have been identified. Inflammatory cytokines such as TNF-Ü, IFN-Þ, IL1, IL-6, and IL-10 evidently play an important role in the pathogenesis of cancer cachexia.In the present study, IL-10 1082 A/G gene promoter polymorphism was detected in Egyptian patients with cancer cachexia and was compared with non-cachectic patients with cancer and investigated the possible role of this polymorphism in the pathogenesis of cancer cachexia.Genotypes of IL-10 1082 A/G were detected by real time PCR in 95 patients with GIT or Lung cancer. The results of the study revealed that IL10-1082 mutant (AG and GG) genotypes were associated with higher risk of developing cachexia in patients with gastrointestinal and lung cancer on comparing cachectic cancer patients and non-cachectic cancer patients with an odds ratio of 5 (95% confidence interval=1.97-12.68 and P 0.001). In conclusion, IL-10 1082 A/G gene polymorphism could be involved in the development of cancer cachexia
