Abstract

Type-2 diabetes mellitus (T2DM) is characterized by increased oxidative stress, apoptosis and inflammation. The current study aimed to investigate the possible beneficial effects of co-administration of biological agents namely, vitamin D (VIT D) and montelukast (MLK) with commonly used anti-diabetic drugs such as; vildagliptin (VILDA), a DPP-4 inhibitor or pioglitazone (PIO), a PPAR-Þ agonist, on metabolic derangements, inflammation, oxidative stress and apoptosis in fructose/streptozotocin (F/STZ) T2DM model in male Wistar rats. Diabetes was induced by maintaining rats on 10% (w/v) fructose in drinking water for nine weeks with a single intraperitoneal injection of sub-diabetogenic dose of STZ (35 mg/kg) by the end of the fourth week. One week after STZ injection, VILDA (10mg/kg/day) and PIO (10mg/kg/day) each alone or in combination with VIT D (500 IU/kg/day) or MLK (10mg/kg/day) were administered orally to F/STZ diabetic rats for the last four weeks of the experiment. By the end of the experiment, blood samples were collected for the assessment of serum parameters including: glucose, insulin, triglycerides (TGs), total cholesterol (TC) levels, as well as, alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities.
