Abstract The fungi A.terreus MH205918 extract showed promising DPPH scavenging activity. Statistical optimization of scavenging activity and total phenolic content resulted in 1.17 fold and 2.37 fold increase compared to basal medium respectively. Butyrolactone I (BL-1) was isolated as major active compound. Total extract and BL-1 showed concentration dependent antioxidant activities in different assays. Molecular docking of BL-1 with the regulatory protein Keap1 showed good interaction. Total extract and BL-1 increased induction of NQO1 in (hepa1c1c7). The total fungal extract and BL-1 showed antimicrobial activity against G+ve and G-ve bacteria. BL-1 showed cytotoxicity against A549 cell line and no activity against normal cell line (BJ1). Two gene fragments, were amplified using PCR from A.terreus MH205918 isolated DNA. One fragment showed 98% similarity to the A.terreus (NIH2624) ATET- 05798 indicated as conserved hypothetical protein in the database while the other, accession number MG881210, showed 96.1 % similarity to the A.terreus (NIH2624) ATEG-00108 gene responsible for Glyoxalase I enzyme |
