Abstract

Highly selective molecularly imprinted polymer (MIP) for solid-phase extraction and determination of Alprazolam in human plasma has been designed and prepared. In order to study the intermolecular interactions in the pre-polymerization mixture and to find the optimal functional monomer in MIP preparation, a computational approach was used.This approach was based on the comparison of the binding interaction and stabilization energy of the complexes between the template and the different functional monomers in different solvents. Based on the results of the computational study, the MIP was synthesized using the monomer of choice, being methacrylic acid, in the solvent of choice, being methanol. Using the MIP prepared, a molecularly imprinted solid-phase extraction procedure followed by high performance liquid chromatography with ultraviolet detection (MISPE-HPLC-UV) was developed for the selective extraction and determination of Alprazolam in human plasma. The prepared MIP possessed a high imprinting efficiency and good adsorption capacity of Alprazolam. For the proposed MISPE-HPLC-UV method, the relationship between responses (peak area) and concentration was found to be linear over the range of 1-100 ng/mL with a regression coefficient (R2) of 0.9992. The proposed extraction technique is simple, effective and can be applicable to the extraction and analysis of alprazolam in complex matrices for therapeutic drug monitoring applications
