Abstract

Prodigiosin is a red pigment produced by Serratia marcescens with reported pharmacological activity. This study aimed to characterize the most lucrative conditions needed to enhance serratia marcescens pigment production, use gamma radiation to obtain prodigiosin hyperproducing strain and detect the presence of two transcription factors regulating prodigiosin production (Hex S and CRP) in seven serratia marcescens strains (MN1-MN-7) and correlate between their detection and pigment production on glucose. The factors affecting prodigiosin production were identified and then optimized through a factorial design of 32 experimental combinations to study the optimal conditions for pigment production using crude glycerol obtained from local biodiesel facility as carbon source for the growth of serratia marcescens. The maximum production (870 unit/cell) was achieved at 22 {u00B0}C, at pH 9 with the addition of 1% (w/v) peptone and 109 cell/ml inoculum size after 6 days of incubation. Gamma radiation at different doses was used to generate hyper pigmented strain (MN200). Dose 200 Gy was capable of doubling the production of the pigment and produce pigment up to 36⁰C. PCR reaction was used to detect transcription regulators Hex S and CRP. HexS was present in all strains except MN1, MN5 and MN200. Transcription regulator CRP was detected for all strains except strain MN3, MN5 and MN200. Only MN5, MN100 and MN200 were able to produce prodigiosin in 4% glucose
