Abstract Aim of work: To investigate the association of GDF5 and ADAM 12 polymorphism and primary (1ry) knee OA in a cohort of Egyptian population. Patients and methods: This study was conducted on 102 primary knee osteoarthritis ( KOA) patients with kellgren Lawrence ( K/L) grading system ({u2265}2 scale)as well as 100 controls. Blood samples (5 ml) were collected for identification of GDF5 gene polymorphism by polymerase chain reaction (PCR) followed by restriction analysis technique RFLP and ADAM-12 Gene (rs1871054) SNPBy (RT-PCR). Results: GDF and ADAM12 (rs1871054) polymorphism were associated with primary KOA in Egyptian patients (P=0.0001). TC genotype of GDF5 presented only in OA patients and was associated with higher grades of OA while CC genotype demonstrated only in controls not in OA patients. T allele in GDF5 [OR=2.58 ( 95%CI = (1.69-3.93) P<0.0001] and C allele in ADAM12 polymorphism associated with higher risk of OA (OR=1.74; 95%CI=1.15 to 2.64, P=0.0035). Conclusion: GDF5 polymorphism and rs1871054 variant within the ADAM12 gene are risk factors for increased KOA susceptibility furthermore, GDF5 polymorphism could be a predictor for KOA disease severity in Egyptian population |
