Abstract

The aim of this study was to investigate the neuroprotective effects of genistein loaded on nanofibers following spinal cord injury in adult female sprague dawley rats (n=75). The animals were divided into five groups: control, sham, SCI, unloaded CS/PVA nanofibers, and nanofibers loaded with genistein. SCI caused a significant decrease in the SOD activity and the level of IL-10 in injured spinal cord tissue. On the other hand the injury induced a significant increase in the levels of NO, MDA and TNF-Ü. Treatment of rats post SCI with CS/PVA unloaded nanofibers improved most of the above mentioned biochemical parameters and shifted them toward the sham group values. The treatment of SCI rats with genistein loaded on nanofibers ameliorated all the investigated biochemical parameters. Genistein induced an increase in the activity of SOD and the level of IL-10, while caused a decrease in the levels of NO, MDA and TNF-Ü in injured spinal cord tissue. However, the present study provides new insight into the anti-inflammatory and anti-oxidative stress mechanisms of genistein action, so further studies are required to clarify the role of genistein and CS/PVA nanofibers in SCI treatment.
