Abstract

The thesis involves the synthesis and characterization of [Pd(AEMP)Cl], where AEMP =2-(2-Aminoethyl)-1-2 methylpyrrolidine. Stoichiometry and stability constants of the complexes formed between various biologically relevant ligands (amino acids, peptides, DNA constituents and dicarboxylic acids) and [Pd(AEMP) (H₂O₂)]²⁺ are investigated at 25{u00BA}C and 0.1 M ionic strength. Complex formation equilibria of binuclear Pd(}}) complexes involving 4,4-bipiperidine and some selected DNA constituent s are studied. The concentration distributions diagrams of the various species formed are evaluated. The kinetics of base hydrolysis of free and coordinated amino acid esters are investigated. The effects of temperature on the kinetics of base hydrolysis of the glycine methyl ester in presence of [Pd(AEMP) (H₂O₂)]²⁺complex are studied. Thermo-gravimetric analysis for Pd(AEMP)Cl₂ is studied. The TGA curves of Pd(AEMP)Cl₂ exhibits two steps, leaving a residue of palladium metal. The X-ray single crystals of [Pd(AEMP)Cl₂] complex is studied. Density functional theory (DFT) calculations have been carried out to investigate the most stable geometries of the ligand and Pd(AEMP)Cl. Ring-opening of [Pd(AEMP) (CBDCA)] and 2 the formation of [Pd(AEMP) (CBDCA-O)(DNA)] is also studied by fitting the potentiometric data for the system consisting of [Pd(AEMP) (H₂O₂)]²⁺, CBDCA and DNA constituents, inosine, inosine-5{u2019}-monophosphate, uracil, uridine and thymine assuming different models. The antimicrobial activity and the antitumor activity were investigated as an application of studying the cytotoxicity of the complex compared to that of the free ligand and finally. The DNA cleavage was studied by mean of agarose gel electrophoresis
