Abstract

Indomethacin is a non-steroidal, anti-inflammatory drug used in the treatment of many diseases but showed many adverse effects especially at ileum. Curcumin is an orange-yellow component of turmeric herb. It has potent antiinflammatory, antioxidant, antimicrobial and anti-cancer effects. This work was performed to study the possible protective and curative effect of curcumin on indomethacin induced ileitis using histological and electron microscopic techniques. This study was carried out on 36 adult male albino rats (160- 200 gm); aged; 12-14 weeks, divided into four main groups; control group: 8 rats, experimental group I: 8 rats received oral aqueous suspension of curcumin 100 mg/kg daily, experimental group II: 10 rats included subgroups (IIA & IIB) received subcutaneous injection of 2 doses of indomethacin. Each dose was 7.5mg/kg separated by 24 hours, and experimental group III: 10 rats included subgroups (IIIA& IIIB) through which ileitis was induced by the same manner as group II then, rats were treated with an oral aqueous suspension of curcumin at a dose of 100 mg/kg daily. This study revealed that: subgroups IIA & IIB showed desquamation, destruction of surface epithelium (ulceration), inflammatory cellular infiltration, subepithelial hemorrhage and statistically decreased villus height at H&E sections. Also, significantly decreased goblet cell number at PAS sections besides electron microscopic changes of the cells lining the ileal mucosa. Subgroups IIIA& IIIB showed marked improvement of the previously mentioned pathological changes. This demonstrate that curcumin administration decreases indomethacin-induced ileitis. So, it could be useful for treatment of intestinal injury. Keywords: Indomethacin; Curcumin; Ileum; Histology; Electron microscopy.
