Abstract In this study, different approaches for micropropagation, in vitro conservation and cryopreservation of grape ”Black Matrouh” and guava ”Al-Baladi”, and in vitro mutagenesis for inducing grape plantlets more tolerant to drought were investigated. Grape shoot tip and nodal explants showed the highest survival% on {u00BE}MS medium with 0.5 mgl-1 BAP, while MS medium with 0.5 or 1.0 mgl-1 BAP produced the highest shoot no./shoot tip or nodal explant, respectively. Established explants cultured on {u00BE}MS medium with (0.75BAP+0.50 IBA) mgl-1 or (1.00BAP+0.30IBA) mgl-1 produced the highest shoot no./explant, while established explants cultured on growth regulator-free MS medium showed the highest shoot length. The highest rooting% was recorded on {u00BD}MS medium with 1.0 mgl-1 IBA and the longest roots on {u00BD}MS medium with 0.5 mgl-1 IBA without significant differences compared to {u00BD}MS medium with 0.5 mgl-1 NAA which exhibited the highest roots no./explant. Guava nodal explants cultured on MS medium with 1.0 mgl-1 BAP showed the highest survival% and shoot no./explant. Meanwhile, explants cultured on growth regulator-free MS or WPM medium failed completely to establish. Established explants cultured on MS medium plus 4.5% sucrose and (0.75BAP+40ADS) mgl-1 or (0.75BAP+80ADS) mgl-1 produced the highest shoots no./explant.
