Colorectal cancer is one of the commonest malignancies worldwide, in Egypt colorectal cancer is one of the most common malignant neoplasms.Colorectal cancer is a multifactorial disease with dietary, lifestyle and environmental exposures on one hand and genetic predispositions on the other hand. In the present study, we examined the hypothesis of whether rs9138 genetic polymorphism in OPN is related to the risk of developing colorectal cancer in Egyptians. This study included 212 subjects; 100 patients of pathologically proven colorectal cancer, recruited from the National Cancer Institute and 112 age and sex matched healthy individuals who served as a control group. The genetic analysis of OPN polymorphism was done using Real Time PCR, OPN plasma level was measured by ELISA.The OPN genotype analysis showed that in female subjects, the frequency of combined genotypes (AC+CC) was significantly higher in CRC patients than that of controls as compared to the frequency of AA genotype (p=0.010). Regarding OPN protein, the level of OPN was significantly higher in CRC patients when compared with healthy control volunteers. (P =0.0001)
