Abstract he present study aimed to detect the relationship between the expression levels of αKlotho and γ-Klotho genes and the development and progression of TC. The study was conducted on 80 individuals (40 TC patients and 40 age and sex-matched. subjects undergoing thyroidectomy for goiter as a control group). Total RNA was extracted from thyroid tissues. Reverse Transcription was done and relative αKlotho and γ-Klotho mRNA expression levels were determined by Real-Time Quantitative PCR. Our study revealed that α-Klotho gene is downregulated in DTC, suggesting that it may act as a tumor suppressor. The α-Klotho cutoff value of ≤0.68 exhibited a significant diagnostic accuracy of DTC with a specificity of 97.5% and a sensitivity of 87.5%. Logistic regression analysis showed that decreased α-Klotho mRNA expression was demonstrated to be significant predictor for the likelihood of DTC on top of goiter. The γ-Klotho gene expression did not associated significantly with the development of DTC. |
