Abstract The present work studies and investigates the in vitro effects of interferon-alpha (Roferon-A) on the replication and cytopathogenic effect of standard strain of herpes simplex virus type 1 (HSV-0. Buffalo green monkey kidney (BGM) cells were used for titration of HSV-land interferon assay. The initial virus titre used in the experiments was 1 x 105 PFU/mL. . Interferon-alpha concentrations used in interferon assay were 3 x 102, J x 103, 3 X 104, and 3 x 105 u/ml. It was found that interferon assay 18 hours before virus adsorption reduced virus titre from 1 x 105 PFU/ml, to a median of 0.15 x 105, 0.2 X 105, 0.3 X 105, and 0.5 x 105 PFU/ml at interferon concentrations of 3 x 105, 3 x 104, 3 x 103, and 3 x 102 u/ml respectively. The inhibitory dose 50 of interferon was noticed at a dose of 3 x 102 u/ml. It was found that interferon assay one hour after virus adsorption by one hour produced a reduction in the initial virus titre from 1 x 105 PFU/ml to a median of 0.25 x 105, 0.3 X 105, 0.5 X 105 , 0.65 X 105 PFU/ml at interferon concentrations of 3 x 105, 3 x 104, 3 x 103, and 3 x 102 u/ml respectively. The inhibitory dose 50 of interferon was noticed at a dose of 3 x 103 u/ml. |
