Abstract

administration of nicotine in different doses for 60 days on fertiltiy in male albino rats . One hundred and ninty four fertile albino rats were used ( 94 males and 188 females ) . The females were used to test lor males’ fertility . The male rats were divided into 4 groups : a control group containing 1 0 rats and three test groups each contains 28 rats . These test groups were given nicotine in three doses ( 0.5 mg / kg b. w. , 0 .75 mg / kg b. w. and I mg / kg b. w. ). Each of the test groups was subdivided into three subgroups , the first was sacrificed immediatly after the 60 days while the second and the third were sacrificed after one and two weeks after stoppage of nicotine administration . Blood samples were collected and used for hormonal assay . The rats were dissected out and subjected to epididymal spermatozoal examination and histopathological examination of their testes . From this work , it could be concluded that nicotine produced testicular damage in the form of seminiferous tubular atrophy , focal interstitium fibrosis and Leydig cell hyperplasia . This tubular atrophy led to decrease in sperm count and motility and increase in abnormal sperms . These effects were dose-related . By the passage of the recovery period , these effects were gradually improved . However , as the dose of nicotine was increased , the residual effects increased and needed more time for recovery Nicotine administration was associated with increased leukocytic count. However , on cessation of nicotine administration this elevated leukocytic count was markedlly improved even with high doses.
