Abstract

The alleles HLA-DRB1 *03011, *1327 HLA-DQA1* 03011, *05011-HLA-DQB1 *0201, *0302 are high risky for IDDM. They are significantly related to GADA, IA2A and CD4. The gene controlling the process of auto-reactive mechanism and GADA production is located within the MHC as indicated by HLA-haplotype concordance and correlations of HLA-haplotypes with the seropositivity and seronegativity of GADA. This genetic control which may form an important portion of the genetic environmental interaction is linked but not closely to HLA as indicated by the recombination fraction ? = 3.802 at 0.01 and ? = 4.650 at 0.05. On the other hand the role of IA2A is less important than that of GADA since the incidence of seropositivity is much less and not linked to HLA. However strong confirmation of this assumption needs extension of this study on bigger number of families including if possible multicase families and for very long time follow up, which should be not less than 10 years to establish strategy for the treatment of the seropositive prediabetics and seronegative sibs with high risk alleles and /or identical haplotypes with the diabetic sibs, before developing the disease.
