Abstract

Sulfonylurea derivatives are still the cornerstone of the treatment of type 2 diabetes mellitus. Their main action is stimulation of beta cells to induce insulin secretion through the closure of the K+-ATP channels. On the other hand, some diabetic patients may be treated with vermicides for prevention or treatment of nemastosis. Mebendazole is a vermicide, which is reported to induce insulin secretion. Therefore, the present investigation was carried out to elucidate the effect of mebendazole on the insulin release and the metabolic action of two hypoglycemic agents; gilbenclamide and gliquidone. Metabolic parameters measured were, liver glycogen content, oral glucose tolerance, blood glucose, and K+, Na+ and Ca+2 levels. The serum insulin level and p-cell function were also determined. In addition, the present work investigated the stimulatory effect of mebendazole on insulin secretion. In this sequence of events, the mebendazole effect on insulin release from normal isolated perfused rat pancreas at substimulatory (5.5 mM) and stimulatory (11.1 mM) glucose levels was studied. The results of this investigation obtained can be summarized as follow: In vivo results: 1. Intraperitonial injection of adult male rats with STZ (65 mg /Kg) and nicotinamide (230 mg/kg) significantly increased blood glucose level and significantly decreased serum insulin level. In addition, STZ induced glucose tolerance and reduced glucose utilization that was indicated by the increase in the area under the oral glucose tolerance.
