Abstract

The study included three main parts: 1- Investigation of the potential preventive effects of curcumin (CMN), resveratrol (RSV), rosiglitazone (RGN), losartan (LOS) and N acetylcysteine on sodium valproate (VPA)-induced liver steatosis in male Sprague-Dawley rats. Results showed that daily administration of VPA for one week to rats resulted in marked microvesicular steatosis with some degree of hepatic necrosis and inflammation. CMN administration with VPA to rats abolished signs of VPA-induced hepatotoxicity while RGN or N-acetylcysteine conferred protection against VPA-induced hepatotoxicity with minimal degree of histological lesions; 2- Study of the potential preventive effects of CMN and RSV on ethinylestradiol and chlorpromazine -induced intrahepatic cholestasis in adult female Sprague-Dawley rats. Results showed that daily administration of EE and CPZ daily for one week resulted in intrahepatic cholestasis. Simultaneous administration of CMN with EE-CPZ produced marked protection against EE-CPZ induced cholestasis; 3- Study of the potential preventive and curative effects of CMN, RSV, imatinib, RGN, LOS and bosentan against Schistosoma mansoni-induced liver fibrosis in mice. Results indicated that S. mansoni infection resulted in hepatic fibrosis development. CMN, RSV or imatinib showed marked antifibrotic activity in decreasing the development of hepatic fibrosis while only CMN or imatinib showed potential beneficial effects in reversing the developed hepatic fibrosis.
