Abstract

The present study was conducted as an attempt to introduce functional genes of adrenal gland and thyroid glands from Naked neck chickens or bacteria (Streptococcus agalactia} through its fragmented purified DNA into the embryonic cells of Japanese quail (Cotumic cotwnix japonica). The effect of these treatments on the ability of hatched chicks to tolerate heat stress conditions was investigated. Hematological and biochemical parameters were evaluated. Moreover, pathological and cytological studies were also carried out. Two thousand eight hundred and eighty eggs of Japanese quail (Coturnix coturnix. japonica} (in three hatches) were used in this study. All eggs were incubated at 37.8°C, in forced ah- incubator. At the 3rd day of incubation, the eggs were divided randomly into 12 experimental groups. Each group of eggs was randomly assigned to one of DNA injection treatments as follows: 1-Adrenal and thyroid ginnds DNA groups: Three groups of eggs were used, for each gland type eggs were carefully drilled and injected with the DNA extracted from adrenal glands of naked neck chickens in doses of 3, 5 or 7 ug/egg (one group of eggs for each dose). 2. Bacteria DNA groups: Further three groups’ of eggs were used and injected with DNA extracted from bacteria {Streptococcus agalactia) adapted for high temperature > 45”C in the same doses applied for adrenal and thyroid groups.
