Abstract

Tllis investigation was carried out at Rice Research and Training Center, Kafr El-Sheikh, Egypt and Genetics Dept., Fac. of Agric., Kafr El-Sheikh, Tanta Univ., during the successive seasons from 1997 to 2000, and aimed to : 1- select and evaluate the best medium could be used in anther culture technique for callus induction and plant regeneration. 2- Utilize anther culture technique for producing near isogenic resistant lines to blast disease for releasing multiline resistant rice varieties. Ten local and introduced rice (Oryza sativa L.) varieties were used in this study. Eight of them were donors for blast resistance genes. These varieties were Pi No. 4, BL,, Zenith, Dular, K3, K53, Akiyutaka and Fukuhikari. All of them were introduced varieties with different blast resistant genes. The two recurrent genotypes were the Egyptian rtce varieties, Giza 159 and Ixeiho. The main results presented in three major sections. 1. selection of the best callus induction medium The results revealed that Fj medium gave the desirable lowest mean value of days to callus formation over all parents (42.39 days). But the highest mean value was obtained from N6 medium (45.26 days). Five parents (Pi 4, K3, KS9, Akiyutaka arid Fukuhikari) gave the desirable lowest mean values of days to callus formation under Fj medium. On contrary, only two parents (BL1 and Ks9) gave the lowest values with N6 medium. I Regasding callus induction trait, Fj medium was more effective for most genotypes than obtained by N6 medium. While, the other callus characters (weight and size) there are no wide differences between both media and the results revealed that callus produced on Fj medium was more embryogenic after transferring to MS regeneration medium.
