Abstract

Diabetes mellitus is one of the most prevalent chronic diseases. Prolonged hyperglycemia in T2DM patients is the major cause of all micro and macrovascular complications including DN which may develop at a later stage of the disease.
Diabetic nephropathy is the leading cause of diabetes-related morbidity and mortality and its prevalence has risen worldwide over the past decades. It has been reported to have an increase in macrophage infiltration and overproduction of leukocyte adhesion molecules in the kidneys.
Engulfment And Cell Motility 1 (ELMO1) is a gene on chromosome 7p14 with 22 exons. ELMO1 encodes for one of the ELMO proteins which promotes phagocytosis and cell migration by interacting with the cytokinesis protein. Researches have proved that the over-expressing ELMO1 contributes to the deposition of extracellular matrix (ECM) proteins by upsetting the metabolism of ECM and diminishing the cell adhesion to ECM, which lead to thickening of glomerular and tubular basement membrane, ultimately resulting in the occurrence and develoment of chronic renal injuries under high glucose concentration.
The aim of this work was to study the association of ELMO1 gene polymorphism and diabetic nephropathy in type 2 diabetic patients.
This study was conducted on 137 subjects (97males and 40 females), their ages ranged between 33-65 years. The subjects in the study were divided into 3 groups:group I( Diabetic with albuminuria) included 47 T2DM patients (31 males and 16 females), their age ranged between 34-57 years.group II ( Diabetic without albuminuria ), included 50 T2DM patients (39 males and 11 females), their age ranged between 33-59 years. group III (Control Group) included 40 apparently healthy individuals, age and gender matched with patients groups.
All Patients and controls included in the study were subjected to the followings: History taking, Clinical examination, Laboratory examination included: FBG and 2h-pp, HbA1c, Total lipid profile(total Cholesterol,TG, HDL-C and LDL-C), BUN and creatinine,Identification of ELMO1 gene polymorphism by Real time PCR.
